A comma makes this two very different sentences.
Despite being believed by many, slugs smiling doesn’t actually prevent you from crying
Or
Despite being believed by many slugs, smiling doesn’t actually prevent you from crying
Slugs smiling would absolutely prevent me from crying. (At least from sadness, it might be so beauriful I cry because of that though)
